The 50WLRSA has the new ratcheting lever drag to prevent it from backing off accidentally and also has the new, larger offset ergonomic power handle. This handle is made out of a non-slip material that doens't get slick when it gets wet. Other than that, no differences.

Can i use the 50 WLRSA for Yellofin Tuna or Smaller Marlin (500 lbs) and what would be the best line set up for this.

Absolutely. As for line, you can go with 100-150 lb. braid or with 80 lb. mono. Either way, you'll have enough capacity and strength to do whatever it is that you wish.
